I was thinking of writting a script for a parody, something like Scary Movie. I know you don't need permission, and i know from Scary Movie 4 Special Features that two jokes cancel each other out. What other rules are there to know

The only rule to a parody is MAKE IT FUNNY!
Use your own imagination to do that!
Life or movie making is not about rules!
Rules hold the imagination back!
Movie Making takes full use of the imagination YOU put into it. Rules would just get in the way as your question confirms. You're so busy wanting to know the 'rules' you don't seem to have anything 'artistic' to say, only technical. The heart of (almost) every movie is the STORY. (exception: Snakes On A Plane...which no one went to see for the story but because there were mf'ing snakes on the mf'ing plane!...lol)...
