How's the fluid level in the transmission? That's the first thing I'd check. If OK then the transmission is probably toast.

I'll assume you're talking about an automatic transmission.....
First thing to do is check transmission fluid. If it's low, then the transmission may not build enough hydraulic pressure to engage. If it's been low for a while, then you've already damaged the transmission. If there's fluid but it's brown and smells burnt, instead of healthy red, the transmission's dead. If you're REAL lucky, you might be OK by topping off the fluid, but if it's already not going into gear, then you're probably out of luck.

Something is broke in the tranny. Have it towed to a transmission shop and they'll inspect it for you>
