42 was the answer a supercomputer gave, when asked the answer to the broad question "Life, the universe and everything".
And since the answer was even more mystifying than having no answer at all, an even more massive computer was developed, which would run for millions of years, to find out the REAL ultimate questiion, to which the answer was known to be 42.
The computer was called "The Earth"
"The meaning of life" question has been asked so often on Q!A, (and no-one ever checks for similar questions, it seems) that 42 seems a hightly appropriate response for a query that seems doomed to be asked and asked and asked.
There's always the Great Green Arkleseizure theory...

I don't know if it's still true, but if you googled "The meaning of life and everything" or something of the sort, google would tell you it's 42.
EDIT: Also, it appears as the Ultimate Question in the "Hitchhiker's Guide to the Galaxy". A computer is asked the question, and after 7.5 million years it replies 42.
