Jules Feiffer has given us a whole gallery of them in Feiffer. Bernard Mergendeiler, his dancer, occasionally, and many others are why he has been celebrated as a poet of Modern Angst.

Charlie Brown was the original stressed out kid, and even Lucy, while described as just a fussbudget (which is different) had her moments.

There are a long line of sad clowns in comics. If I'm not mistaken, even Charlie Chaplin had his own strip, which Elzie Crisler Segar worked on before creating Thimble Theater Starring Popeye. And of course Frederick Burr Opper's Happy Hooligan was named ironically.

And of course Krazy Kat's Ignatz Mouse not only had a problem with Katz but strongly resented being the object of affection of someone of indeterminate gender. His plight is handled with considerable sympathy and sophistication in the original George Herriman comics.
