I'm not sure, medications usually aren't themselves, but they are detected by antibodies which are proteins and that starts the immune response that rejects the medication. I guess you could call an antibody a marker protein, though.

The surface area would increase as the volume increases, but the ratio of surface area to volume would decrease.

Most proteins are made in the cytoplasm or ER.

Proteins already have all their amino acids by the time they reach the Golgi
