They both come from the family Bovidae, but that is where how they are in common more or less stops.

Well to start bison live in North America and Buffalo live in Africa and Asia (cape and water).

Bison have a large shoulder hump, buffalo do not.
Bison have more hair.
Buffalo horns are connected to their skull, bison are not.

The reason people goof in calling Bison a Buffalo is because when settlers first came here they thought that bison looked like a type of buffalo... it just stuck.

Strictly speaking, "bison" is the term for the New World temperate indigenous cattle. The European "wisent" is similar. Buffalo are the wide-horned southern Asian and African cattle.
