That's NOT from the Constitution. It's from the Declaration of Independence. Jefferson was looking for a way to claim an authority for the actions they were about to take that was independent of the the British.

In context of the "The Declaration of Independence", it means that the truths mentioned afterwards need no proof [self-evident] because they are granted from God.

"...that all men are created equal, that they are endowed by their Creator with certain unalienable Rights, that among these are Life, Liberty, and the pursuit of Happiness"
