How i can differentiat and identify different ports for cards such as ISA and PCI.?

Well the connector configuration is totally different.

The PCI connector on the card is only about 3 inches long.

The ISA connector on a card is about 5 inches long.

You simply cannot mistake the two sizes and they certainly are not interchangeable.
