There's a number of ways to tell, but none of them are 100% guaranteed if you don't have proper measuring equipment. For one, diamonds disperse light -- if you put it in front of a piece of newspaper, you should not be able to read through the diamond. Also, try fogging up the clean stone with your breath. If it is a zircon, then it will hold fog for at least 2-4 seconds, whereas the diamond will disperse the heat almost immediately.
You can also try the old hitting the diamond with a hammer test. Watch out though, because a zircon still may not break, whereas a diamond may in fact break!

Due to this there are imperfections in the carbon. A real stone will be perfect - absolutely perfect. Even there are synthetic diamonds which is not identifiable. Its better to take it to an expert. 2) Sandpaper test. since diamonds are one of the world's hardest stone and it won't be scratched by the rough surface. If you scratch sandpaper to a real diamond then nothing will happen but if it's a cubic zirconium, it will scratch it up. 3)Fog Test. Breathe hot air on your diamond as you would do on a mirror. A fake diamond will fog up for a short period of time whereas a real diamond will not because it won't retain the heat

A jeweler will pay you the wholesale value for the stone.
Another way is to do hardness testing (diamonds are the hardest known natural substances, though some man-made minerals are coming close).
You could also test the diamond by x-ray diffraction and compare the pattern generated with that expected of a diamond.
You could completely crush the diamond and see if you wind up with pure carbon (graphite), anything else means that you had a fake....of course I wouldn't recommend this with what looks like a gem quality diamond or one that is large enough to be commercially valuable.
But, the most certain way to determine whether it is real is to give it to her and see if her eyes light up. If so, you've got a real gem. ;^)

Real diamonds can cut glass ... other than that - I couldn't say for sure. Some of those fakes these days can be very deceiving! If your buying on-line do your research first. Check out the company and the products they sell. Pay with a credit card - you can dispute a purchase if you run into problems with it being fake.

well are you talking about fake (IE..CZ) or are you talking about synthetic (cultured diamonds) if its a zirconium then run it down your neigbors windsheild.. however if it is synthetic.. IE gemesis or adia or any of the others.. there realy is no way of tellin the diff.. BECAUSE a synthtic diamond is the same thing as a mined diamond.. in many cases when a mined and a synthetic where placed in front of a gemist they had trouble tellin tthem apart... however many chemists can tell the diffrence by the way they carbon atoms are formed.. the age of the diamond.. older diamonds look diffrent then newer diamonds.. think about it the youngest mined diamond is 900 million years old... and we now have companys that are making diamonds in 3 days... but the real way to tell them apart is by the AIG and CGA certifications.. if it didnt come with a cert then its prob fake!! even synthetic diamonds come with certs stating cultured diamonds...

diamonds cut glass.
try the dot test. draw a dot on paper, put the gemstone on top of the dot. if the dot comes up solid and you can easily see it its fake. if the dot appears smudged its real, because diamonds distort the light.
put it under UV light. if it is real it should be glowing bright blue.

Petrographic microscope and a (Dana) manual with refractive indices.
Many minerals ( even some clear ones) will scratch glass eg Quartz !
