Do I have to buy the first one to fully comprehend it or can I buy the newer one, Nightfall.

Depends on what kind of game you want to play. The only connection between the stories of the games are between Prophecies and Eyes of the North. Otherwise each game is a complete stand alone, and if you start a new character in any one of them you are tutored as if you haven't played any of them at all.

Prophecies: Is the first. It has the slowest leveling, and the longest story quest. There are only 6 basic classes, and nothing about it other than the story and a few skills are unique to it alone. It is however, the one most closely liked to Eyes of the North, and you could care about the characters involved more if you play it before getting GW:EN.

Factions: Is the version of PvP. Otherwise I haven't really played it and can't say anything else about it. Has the assassin and ritualist classes.

Nightfall: Has heroes (improvable henchmen) which makes soloing a LOT easier. Nightfall also gets you to level 20 VERY quickly with very high EXP quests and missions. Has the dervish and paragon classes.

I think the best place to start is the "Prophecies" campaign. It is the original and will give you all of the back story.

If you plan to do a lot of solo playing (with computer-controlled party members to assist you rather than other online players), you might want to start with the "Nightfall" Campaign. It includes smarter party AI (called Heroes) that will make playing through teh game by yourself a lot easier.

If you enjoy PVP the most, you would do well to start with the Factions campaign. It is a very pvp-centric one.
