There isn't a best class in Guild Wars. Strategies in PvP and PvE are always changing, so the most effective class is always changing too. Skills equipped matter more than clas, class just determines what skills you are likely to have.
Play the class you like the most and try them all, maybe you'll find your own combination.
I've heard some whoopers though: like an Elementalist/Hammer Warrior, and a Ranger/Healing Monk.

The game is fairly balanced. For PvP there is no best class. Just choose the profession you enjoy playing. I'd recommend trying at least 4-5 different ones.
For PvE it's the same. If you are going to be doing a lot of pugging though, it's easier to get into groups as an Ele, Monk or Necro.
