When the men went to fight in WWII, the women had to take their jobs in the factories. This encourages women to go out and work, supporting the United States and its economy at a time of crisis. This has undertones of feminism.

The answers you have gotten are correct in that it refers to women being able to do the job required to win the war, but I think the "we" goes beyond gender.
It also speaks to the circumstances the entire nation was in. We had been sort of "clotheslined" by the attack at Pearl Harbor (an illegal football move, where a defender sticks out an arm just when the ball carrier comes by, hitting him in the throat, I used it here because the ball carrier knows someone is going to hit him, but a hit like this is one you don't look for) and the whole country was woefully unprepared.
The "we" here is the nation as a whole, who are going to come through, as the President said and "win through to ultimate victory, so help us God".

The woman in the poster is "Rosie the Riveter". She represents all the women who went to work during World War 2 to replace the men in the jobs they left vacant to go to fight in the war. Many women became riveters in the ship yards to build battle ships and such to use in the war. Many other women also filled in other jobs where the men usually worked to keep the businesses going during the war. Before that most women who were married stayed home and raised the children. They did not work outside the home as a rule.
