My question to you is, can you install the old glass handles 2 inches lower or higher than the original hole? If so you can install the handles now and then find some wood the same size or a little thinner and use a hole saw to cut out a plug. Once that is done get a fast drying epoxy and cement the plug in the hole. Once dry, fill the whole in with wood putty and let dry. Sand and paint. If you have to use the same hole, the repair will have to be done before installing the glass handles and locks

A typical doorknob hole is 2 1/8". The residential center setback is typically 2 3/8 inches or sometimes 2 3/4 inches.

The most common cross-bore dimension diameters are 2 1/8" x 1", with a back set of 1 3/4" or 1 3/8" [ door edge to centre of large hole ]. If your hole is larger, a pair of " scar plates " or a " wrap around " can be used to make the door serviceable.
