It will not allow fuel to go into the carbs. How do I release the vapor lock? Do I need to take it out of the bike?

Remove the fuel line from the petcock and turn the petcock to run - gas should flow out.
If not - remove the gas cap, make sure there is gas in the tank, make sure the petcock works (connect the fuel line, turn on the petcock and blow through the line).

Install the fuel line and disconnect it from the carb - gas should flow out of the line.
If not - all of the above, or clogged line (blow through it).

Open the drain screw on the carb and install the fuel line on the carb - gas should flow out of the carb's drain.
If not - all of the above, stuck float needle, stuck float, clogged drain (clean the carb).

try some additive in your gas. There is one called gas dri that works sometimes. If that won't work replace the fuel line. Maybe try routing it a different way, sometimes it's not vapor lock but the fuel line is too close to a major heat source, if you;ve changed exhaust or other parts it could be this boiling the fuel in the line. If there is a fuel filter check to see that it's not clogged as well.

If it really is vapor lock, then there is only one fix: Change your gas cap. The cap is supposed to allow air in to take the place of the gas that leaves the tank. If it doesn't then the gas won't flow: vapor lock.

I've been doing this for years and never seen a vapor lock on a bike, my first suspect would be the petcock filter sock plugged up (dirty) remove and clean then try again
