No, James the Just, the first leader of the Christian Church after Jesus' death is supposed to have been Jesus' half brother. Apparently he was the son of Joseph by a previous marriage.

It depends on what John you're talking about. John the Baptist was Jesus' cousin. The other John was His disciple.

To the best of my biblical knowledge, he is not. He did have a half brother I believe, James.
