Henry did have a sister, Margaret, but she didn't marry the King of Portugal, or murder him.
The series merged his two sisters into one character, and called her Margaret.
The younger sister, Mary Rose (Henry's favourite), was a very pretty princess. She married the elderly King of France, on condition that when he died, she could marry the man of her choice. This was, indeed, Charles Brandon.
The series does not follow history, and is not to be recommended for people who want a truthful version, though it's fun to watch if you can overlook the inconsistencies and anachronisms.
Here's the truth about Mary Rose:

As Lili already answered, it was Mary (Henry VIII's youngest sister) , not Margaret Tudor, that was married to Charles Brandon. Mary Tudor was married to the King of France. After her husband died, Mary returned to England and fell in love with Brandon. Even if it was considered an unequal match (a Dowager Queen and a non-royal nobleman), Henry VIII granted permition for their marriage, due to his affection to his sister. Contrary to the TV show, their marriage seemed to be a very happy one. Mary's granddaughter was Lady Jane Grey, who inherited the throne for only nine days and was finally beheaded. The real Margaret Tudor (the eldest sister of Henry VIII) was married to the King of Scotland instead. Her granddaughter was Mary, Queen of Scots.which had a similar end with her cousin, Lady Jane Grey.

No, this has already been asked and it appears that the makers of the programme decided to invent a sister who was a blend of Margaret, who became Queen of Scotland and Mary, who became Queen of France. Portugal doesn't come into the picture at all, any more than does the smothering episode. The makers of the series had a vivid imagination. It's a pity they couldn't find a redhead to play the King: I have to keep reminding myself who the actor is meant to be! http://womenshistory.about.com/od/tudor/a/tudor_women_2.htm

l am afraid you are on very uncertain ground if you use this programme to learn Tudor history. Indeed, you will need to know a good deal about this period to follow the programme, which l find very disappointing and not up to the BBC usual standard. That said it was Henry's younger sister Mary who made the deal that if Henry chose her first husband then she should choose the second herself. This what happened and far as we know she lived happily ever after. She certainly did not- as far as we know - murder her first husband.

Well they seem to have a massive dose of poetic licence. I watched the first in the series of what I expected to be an interesting historical drama, was in fact just a big sex romp. Flippin eck tis enough everyone on yahoo wants to talk sex, without what I thought was a bit of good telly making you feel inadequate as well. And they didn't even have channel 4 in those days.
Can I just say that the bloke who plays Henry was fantastic in Gormanghast. You never ever saw his BTM in that. Thank goodness.

Henry VIII did have a sister called Margaret but she was married to James IV of Scotland, and mother of seven children, one of whom became James V of Scotland.
No murder as far as I can tell.

no she did not!!! Margaret Tudor was the eldest of Henry VII children, she was married to the King of Scotland James IV. She was the grandmother of both Mary Queen of Scots and Mary's husband Lord Darnley ( through her daughter Margaret Douglas, from her second marriage to Archibald Douglas) she had nothing to do with the King of Portugal.
the Tudors is a Programme my wife watches, I personally find the actor chosen to play Henry a complete Joke?
if you want more info - like the other Margaret's descendants the Duke of lennox and Arabella of whom had a strong claim to the throne, she married Edward Seymour himself linked to the throne through the Grey family, descended from Henry VIII younger Sister Mary. just e-mail me I would be happy to help.
EDIT: my wife has just told me that in the programme she later marries the Duke of Suffolk. this is WRONG!!!!!! and I don't really know how they can do this on tv.
the sister of Henry VIII that Married the Duke of Suffolk was......... Mary! she was married first to the King of France, Louis XII, she despised him, but it is said she - made love to him to death(the nice version lol) Henry sent the Viscount Lisle(Suffolk) to bring his sister back and try and convince her to let him marry her off again, this time to the Emperor Charles V, but this was against the personal deal she had made with Henry. She pleaded with Charles Brandon (Lisle) to marry her, for they had a history, he at first said no, but she must have been very convincing, for they eloped. Henry, after initially hitting the roof, accepted the marriage, made Brandon the Duke of Suffolk and sent them both off to Suffolk to live happily ever after. Their Granchild was, The Nine Day Queen, Jane Grey! their descendants married into the Lennox's the descendants of Margaret Tudor and doubled their rite to the throne of James I Stewart's Britian!
e-mail for more info if you like

It was probably both true, and untrue. I'm not sure, but it was conspiracy I'm sure. Sift through the Wiki and a few sites on Portugal royalty and the Tudor Dynasty.
