The layout for the transmission is:-
P, R, N, D, 1, 2 and sometimes 3.
Normally you would be in a drive, but if you are crawling along in traffic (Toronto/Calgary in the rush hour). You would shift to 2 to prevent damage to the trans. If you are driving and come to a steep hill, you would use one to keep the vehicle from running away (basically it is held back). Again you would not stop at the top of the hill and then go into 1.
So the answer is No, you would not hurt it!!!

depends on how your driving it. if your standing on the accelerator all the time while it is in 1st, it will put un-needed ware on the 1st gear clutch packs. That transmission was designed for economy not for racing. doing quick starts all the time rather it is locked in 1st or in drive will quickly reduce the life span of the transmission. you will also run the risk of over revving the motor (yes even with a rev limiter) if you want a car with more power, sell the Nissan and pick up something else. or spend some time and money, modify what you have to handle the added abuse. just keep in mind that everything in a car wares faster the harder you use it.

The 1 on the shifter has nothing to do with taking off. Yoy are in 1st gear when you stop and moving the shifter is an imaginary thing. The 1 and 2 are locks that hold the tranny in that gear and changes the shift point to a higher rpm for going up steep hills or pulling a trailer.

if your starting off in first gear and slaming in to drive to get the kick at a high rpm yes it will eventually damage it,, the transmission on that car i believe is the side mounted ,and a front wheel drive,

yup because the other gears are 4 other uses unless u have one of those new trans wich have the sport shifter from automatic to manual dont do it i burned my transmission and it was NOT cheap ! belive me

No it won't hurt it. However it will be about the same launch from drive. If you have an overdrive button turn it off and leave it in drive. That will be the best bet.

No it should do no harm. You will just wear the car out quicker by always being into throttle,don't over rev engine
