PS I know the rules of sudoku; but I refer to the random shaded squares that always appear, they MUST have some significance?

Do you have an example?

Each of the 3 x 3 grids are kind of blocked off and sometimes shaded, to signify that that 3 x 3 grid needs to have 1-9 in it as well. So not only do you need the row to have 1-9, the column to have 1-9, and the 3x3 grid to have 1-9, none of thsoe can be duplicated in any given row, column, or grid.
