I would personally do something like hemophilia, Down syndrome, Cystic fibrosis, or Sickle-cell anemia.

Down's Syndrome - there's something in Yahoo news today about how treadmills may help those afflicted with it to walk.

Taysachs would be one that most people woul dnot already have chosen; or your could try sickle cell anemia.
