Absolutely. As the door accelerates from a set position, the fly will impart a force, albeit a small one, on the door. The door will impart an equal and opposite force on the fly. This force is proportional to the acceleration of the door, so the fly is certain to notice a door opening more quickly.

This force will translate into an increase in pressure between the door and where the flies legs contact the door which is equal to the force divided by the this contact area.

Since this contact area is very small, it will magnify the effect of the force on the pressure. The pressure difference will still be very small, but it is unlikely that it would be unnoticed by the fly.

I suspect the fly knew the door was not moving when he landed, thus he stopped beating his wings, the fight or flight response would make him fly off when his sense of balance detected the movement of the door, a good question would be does the door move when the fly lands on it?

It depends on hoe fast/ gently you open the door!!!! just like the earth is also moving but we do not feel it because the huge mas is moving constantly slowly.
