Aspergillus niger is a type of fungi. it's cell wall is made up of chitin. these chitin wall have N-acetyl glucosamine linkages.so, how to break chitin wall &get the enzyme-Glucose oxidase.

Err.....freeze thawing method, (or) then using bead homogenization (with controlled temperature below 0).
After that use a temperature controlled ultra-centrifugation.

The temperature must be keep as low as possible to avoid denaturation in your target enzymes. Also, remember to control pH level in the solution cause enzymes are very sensitive to pH.

That's by physical way....for chemical way....err...i'm not sure about that.....(cause i haven't done it before) and its vary depend on what enzyme you want to extract.

Freeze the mycelia in liquid nitrogen, and then pound them into powder in mortar and pestle.
