Tough Call!!! I am a huge fan of both Chris Rock and Dane Cook!! I think each of their Stand-Up is "the funniest" but in their own way... If that makes any sense.....

Rowan Atkinson - Blackadder is hilarious (and if you're in love with Hugh Laurie as House, check him out in season 3 and 4 to see what he does best), as is Mr Bean and his stand up stuff is better than both.
Close second would be a tie between Lee Mack (English), Danny Bhoy (Scottish) and Chris Addison (English).
Its something about the British sense of humour (Monty Python is awsome too), plus the accent just makes everything sound funnier.
