Here's the recipe for 50X TAE buffer
first add about 900ml dH2O
242 g Tris Base
57.1 ml acetic acid
18.6 g EDTA
Adjust pH to 8.1
Add dH2O to 1000ml

i only added 121 g of Tris but i already have 1L total. how do i fix this? thanks.

2 ways:
1) make 2 or 1.5 L and don't mess up again

2) take some time using algebra and calculate the ratio's and add in what else you need to equalize it

Most of us have done that sort of thing at one time or other. I would just add enough T, A and E for 1.5L and adjust the volume to that. Incidentally, that's why I usually start with about half the volume of water (500 mL in this case) to make these solutions - that way, when I realize I've fouled up, I can correct it with less trouble...

Just add the rest of the Tris, it won't make that much of a difference in total volume :)

Or you could make 25X TAE instead by adding ddw to 2000ml :p
