The Spruce Goose was the biggest for sure, but it was only flown once for a few minutes, so I wouldn't really count it. I believe the largest flying boat to actually see regular service at this time was the Mars used by the US. It was envisioned as a fast troop transport to ferry troops across the pacific. But by the time it was in service the idea turned out to be unnecessary.

It was the "Spruce Goose" H-4, HK-1 produced by Howard Hughes. It was a prototype and flew only once after the war in 1947.

The china clipper.navy named it P B Y. It was built later on a larger scale armed with two 30 caliber
machines shot out of the bubbles on the sides.
Later it was used for air sea rescue down
pilots or PT boats damaged so bad they to go in the sea,
THE BIGGEST WAS THE HOWARD HUES
SPRUCE GOOSE. but didn't get finished till after the was was over.IT only flew once,three ft
above the water for several miles.it taxed back and never flew again.
