I think probably the best way to answer this is to go to the site Superman Through the Ages and read the on line comic The Luthor Nobody Knows. It tells about how they knew each other as teens and what happened to their friendship. It is a sad, proud and painful story, and, I think, it teaches us all a warning lesson about how to act toward people who may be jealous of us.

My advise is to not go off Smallville for Superman backstory. Clark and Lex were friends and then had a huge falling out.

First season: Clark saves lex life and they become friends. Lex become protective over Clark, but hides a certain obsession towards him, since he feels Clark might have done more than 'save him from the river'. Lex wants to do good, but his reputation usually precedes him.
Season 2: Everything goes well. Lex is still trying to find Clark's secret on the sideline, but still isn't bad. He tries his best not to be like his father, Lionel, which at that moment, was the bad Luthor. Lex's life has been attempted many times, but ultimate one is his newly wife, Helen, who made his plane crash in the ocean.
Seaon 3: Luckily, Lex crashed on an Island and somehow survived. He comes back to Smallville after being rescued. Seek a revenge on Helen, but doesn't kill her. We learn more from his past, for example, he had a younger brother who died. Lex remains good, maybe getting off track few times, but nothing major at that point. Become crazy at one point, but was actually poisonned by his father. His affection for Lana grows slowly. Lex is good at this point. He makes his father go to prison and protects Chloe Sullivan. At the end of the season, Clark discovers Lex's secret room which contains stuff on Clark and the accident of season 1. The friendship is over.
Season 4: He has an obsession for some mysterious stones. Lana comes back from Paris. her boyfriend soons follow. Lex is a little more dark, but still not evil. He makes sure Jason loses his job at Football assistant. He clearly seems to like Lana. Becomes friends again with Clark. Lex's dark side seems to take a little over at the end of season 4 when he forces Chloe to go with him at the caves, when he realizes his stone has been stolen.
Season 5: Lana and Lex grows closer, despite she dates Clark. He hires a woman to make Lana and Clark break up. He runs for Kansas Senator and hires someone to bring Jonathan kent down. His friendship with Clark will break once and for all. later, he gets the girl. Lex is certainly darker here. Evil? Hmm, maybe a little.. He did captured Aquaman and made him deshydrate himself. At the end of the season, Lex becomes possessed.
Season 6: Lex gets Lana impregnanted (fake pregnancy to get her to marry him). We learn he as a secret facility (33.1) where meteor freaks are held. He abducts people to take a sample from them (Chloe has been abducted). He is forming an army of super freaks. Yeah. Lex is definitively evil at this point. At the end, he slaps Lana (joyful moment) and later gets arrested for her murder.
Season 7: He was drowning in the police car (Epi 1) and got saved by Kara. From there, he seems to have changed. He wants to be good again. Will it work? Of course not.. He is not meant to be good. But we'll see how it goes.
***When we were friends**: As I stated above, they used to be friends, but few events grew them apart.
Personnally.. Lex is my favorite character. MR is the best Lex Luthor ever. I like to see how Lex comes evil... hmmmm..
But this is the Smallville explanation. Comis, movies.. not the same... But in the end, Lex will always be evil. It,s his destiny.

I'm currently on season 6, & he became pretty evil in season 5, though not completely evil.
