Factual questions should have correct answers; details included help tiebreak.
Opinion questions should be clever, witty, or creative; I either get a laff, or glean a new or subtle insight. Since there is rarely more than one or two such answers, selection tends to be EZ.
That's me, however. Many agendists simply want their own biased, myopic viewpoint validated, and pick the biggest woof. So it goes.

Unfortunately, what Chipmaker says is correct - too many times, people just want someone to say, "You're right, I agree."
I do prefer an answer that reflects at least slightly my own beliefs, but I'm open to any opinions. Sometimes a person will see things completely differently than I do, but if their argument is clear and strong, then I may still give them the vote for best answer.
If it's an opinion question, I also like to see someone make the effort to answer in more than 10 words.

It depends usually I pick the answer that I like the best or that I think answers my question the best as in that it answers every part of my question even the added questions in the added details and how they go about bringing their point across whether I agree with it or not is a whole different thing altogether.

I base the best answer on how well the person answers my question and how it helps me. How informative it is and to the point. I think once you read all the answers the best answer will just naturally come to you.

I prefer creativity and not someone just looking an answer up on the internet and typing it down. I want to hear an angle that I have not heard, a little humor is good, and I want to see people thinking.

The best answer of the answers. For the record, I am NOT retiring. I was never going to retire. That was b.s. I just wanted to see what people really thought. I am having fun here and I am going anywhere. I couldn't retire when there is still questions to answer, questions to ask, points to score and to make Red Sox and other people that don't like me upset.

I've noticed most people choose their best answer based on whoever agreed with them the most. I choose the one that makes the most sense to me, whether I agree or disagree.

The best and correct answer, obviously. Ones with detail and flair, be it humor, links or background, get my attention quicker.
