WHILE I was growing up, (comma) my parents were always very protective of MY YOUNGER BROTHER AND ME.

Don't listen to anyone who is having you say "of my younger brother and I" because they're wrong. "My younger brother and I" would only come at the beginning of a sentence where the pronoun would be used in the subjective case, therefore warranting a subject pronoun (in this case, the word "I."). The pronoun you're looking for is objective, therefore you must use the objective case "me." However, the actual noun (in this case "my younger brother") always comes before the pronoun (in this case "me).

I think it's already correct but you could try this:

My parents were always very protective of me and my younger brother as I was growing up.
