Off the top of my head, it's a 3-way tie:
1. Mickey Rourke in the movie "Barfly," meeting with a literary agent who's discovered his writing and tells him, "Don't you understand? You've been discovered!"

One of my favorite movie quotes is from the movie "A Fish Called Wanda" which is absolutely hysterical with Jamie Lee Curtis and Kevin Kline.
