I have checked the spark plug, the fuel line and plugs and there are all fine. What could it be?

cheapest and easiest way for diagnostic: Try starting fluid in the intake right at the air box. If if fires up for a second and won't stay running, more than likely fuel problem.
I know you checked the plug but check for spark too. take plug out connect the wire. with some insulated pliers hold end of plug against metal part of quad (engine works fine) attempt to start and see if you have spark.

ok i no this may sound silly but if you have checked your petrol tank and there is a bit in there, you may not no but there may not be enough as it has to get over this small little mound.
check your oil

having a stuffed chain can do it

wash your air filter put it in mineral turps then warm soapy water then fresh
if you have a crap airfilter that can do it
take off your air filter spray some wd-40 downn there
unstick any dirt that has gotten down there

otherwise to the moto shop it is to get a good old check up!!

Always give year, make, engine size and model # when asking questions. We need all of that info to give an informed answer.
2 stroke & 4 stroke engines can have similar problems with different repair procedures.
