I have recently booked a 7 night stay at the Quality Inn in downtown Los Angeles. I booked via Lastminute.com, I have scince had a change in circumstances and now need to stay a full 7 days earlier, if available, what are the normal rules about amending the hotel ? I have contacted Lastminute.com and are still awaiting a response. I am looking to travel 14th - 20th November.

If your reservation is confirmed, just call the hotel. Explain the reasoning and ask if it can be changed. This is the problem with last minute reservations, in that you are reserving "space available". Should there NOT be space available 7 days earlier, then you may be out of luck. Either way, you should call direct to the reservations at the hotel itself.
