The above answerers are correct, but the details they don't add is that the Turtles were originally intended as a spoof of Daredevil. There was a little boy carrying a bowl containing four little pet turtles from a pet store when a truck carrying containers of radioactive waste almost ran over a blind man crossing the street.
A teenage boy saw that the blind man did not notice the truck and tackled him out of the way. That is exactly how Matt Murdoch (Daredevil) lost his sight and got his powers, when a can of the radioactive waste broke open and got in his eyes.
In the original TMNT comics, the teen was not shown getting hit with the radioactive waste. Another container fell off the truck, bounced up and knocked the fishbowl with the turtles in it out of the little kid's hands. The glass broke and they went right down the storm drain, since he was standing right over it. So did the radioactive waste, because that cannister also went down.
Splinter was already down there, having escaped when the Shredder murdered his owner. The goo mutated Splinter, too, making him smart enough to remember all the ninjs training that he used to watch his owner do.
So when he found the Turtles had mutated too, he taught them the ninja arts and gave them names from a book of Renaissance art that washed down the storm drain one day.
It all started out as a spoof.

I originally wrote the Ninja Turtles in grade school and Kevin saw what I made as comic book. We did it for fun and he actually made it really work! It started when my turtles died and had to flush them but then I thought maybe they didn’t die and became super heroes! Made me feel better as a kid and now he has made so many others feel good. Originally they where Bernie and Bernadette.

the green ooze transformed them, and master splinter. Splinter had been the pet rat of a ninja master and he taught the transformed turtles the ninja moves he had seen when he was a pet.

somebody threw them out or flushed them down the toilet into the sewer where there was some kind of toxic waste that they walked through. Splinter found them there and he also walked through it.
