My rosey red minnows are very active there is one large one that stays in the rock cave and it fights off other fish that come close. I think it is a male but im not sure because it has no dark spots or anything. Im pretty sure out of the other three their is one female is there a chance that they might be spawning?

There's a very good chance their mating. Fathead minnows (what rosy reds are) are cave spawners with the male guarding the territory /nest. Just keep everyone well fed and you should be able to keep the fry.

Males are SMALLER then the females, and not as fat.
