I have loaned out all my HP books, it's been some time since I read them, but this is what I have for an answer: They each defied Voldemort while protecting Harry, and of course his mother did with the most effect because of her bravery and love... they appeared to Harry in moments of great stress, too, in their spirit kind of way, again to protect and guide him.

Remember in HP7 how they and the others who loved Harry appeared to help him walk on ahead to Voldemort to ultimately defeat him? So there are two for sure, and I seem to remember a third time, perhaps the words spoken in the Mirror of Desire... or those times they appeared at his side in other times of stress. And that stag patronus!
Anyway, I'm sure others will log on to this question and clear up my untidy answer. Good luck!

Lily Potter, Harry's mother, is a Muggle-born or Mudblood. James Potter, Harry's father, is a pureblood - just as much as any Black or Malfoy. However, he was probably considered a blood traitor after marrying Lily. Voldemort placed no importance on Lily's life, with her being Muggle-born, and little on James'. More importantly, however, he believed due to the prophecy that he had to kill Harry or be killed himself, which worried him more than killing a couple people, even if they had both been purebloods. So Harry is a half-blood, because a wizard is only considered a pureblood if all four of their grandparents and both parents were wizards, and even then not so by the more stereotyping familes like the Malfoys.

The prophecy means that James and Lily had somehow defied Voldemort three times BEFORE Harry's birth. Their activities are never spelled out, but we can probably assume pretty confidently that they were activities conducted as part of their work in the Order of the Phoenix.

JKR did not pan out the books to clarify how exactly Harry's parents had thrice defied Voldemort. However, since they were in the Order and fighting the rise of Voldemort, there must have been 3 separate occasions when they escaped him.

I don't think the time that Voldemort goes to Godric's Hollow counts as one of the times when they defied him because:
1. They could not defy him then - they actually fell victim
2. Voldemort chose to attack Harry based on the prophecy, which means that his parents would have already defied him thrice before he set out to kill them finally

yeah ... harry's parents defied voldermort thrice on previous occasions wen they wer a part of the order..they hd escaped from him thrice..so dats wat it means..they defied lord voldemort before harry's birth and so did neville's parents..so dat was the confusion..to was it neville or harry!!they never mentione the ways they defied but...

We do not know for certain. What is implied is that as members of the Order of the Phoenix, they confronted Voldemort three times and lived to tel the tale.

Later, Dumbledore apologizes to Harry for withholding information over the past five years. He reveals the lost prophecy, which was originally given to him by Sybill Trelawney. Either Harry or Voldemort "must die at the hands of the other, for neither can live while the other survives". Dumbledore also reveals that, due to when the boy was predicted to be born, Neville Longbottom could also have been the child in the prophecy. Dumbledore believes Voldemort chose to attack Harry because he is a half-blood like himself; Neville is a pureblood. In so doing, the Dark Lord marked Harry as his equal. Dumbledore also discloses why he continues to send Harry back to the Dursleys' home for the summer. He tells Harry that when his mother died to protect him, this initiated an ancient magic. As long as Harry stays at the house of his blood-relative long enough to call it a home, it would provide for him a shield protection not even Voldemort is able to overcome. Furthermore, Dumbledore explains to Harry why he had not made him a prefect: he thought that Harry had enough to worry about and did not want to burden him with more responsibilities.

Nothing is ever mentioned in the books about Lily and James defying Tom Riddle.Never.
