I'm not afraid of ghosts would imply a general bravery in the face of ghosts. I'm not afraid of ghost is grammatically incorrect, because you can't use a noun like that without an article - either "the" or "a". I'm not afraid of the ghost would suggest that there is a particular ghost of which you are not afraid. I'm not afraid of a ghost suggests that no individual ghost could scare you (although presumably multiple ghosts at once could).

It would be I'm not afraid of ghosts. Um, ones plural and ones not. If you wanted the singular, it would be I'm not afraid of a ghost.
