depending on the expression and intones it could mean a couple of things.

do you mind doing this for me?
do you mind if we join you?
do you mind giving us a minute?
these are in the context of polite requests.

now,

do you mind?! stop that!
do you mind?! that's my seat!
do you mind?! i was here first!

these are not polite but rather aggressive.

if some one asks if you mind, it can depend on the situation and tone of voice.

They can be asking if you care if something is done...like "do you mind if I sit down here?" they just want to know if it's ok

if you do something that irritates someone on the other hand they could sarcastically say "Do you mind?!"

generally it means "would it bother you if I did this" or people say it when they want another person to stop something they are doing....the english launguage is funny that way its got all kinds of double means to words and phrases

also known as someone asking you for a favor, maybe for you to mind your own business or to leave that person alone.
