In the system tray there is usually a little printer icon.
If you double click it is should show all the things that are sent to the printer for printing.
You can right click these and choose 'Cancel'.
If you can't get this done fast enough, remove the paper for the printer, then do that.

If you have accidentally started a long print run (e.g. you meant to print one page but you asked for a whole book) the first thing to do is remove the paper from the tray or remove the tray. Now you have time to go into Control Panel, select Printers and other hardware, then select Printers and faxes. Select your printer, Cancet the print job, then turn the printer off, replace the paper and turn it back on and start again.

Most printers have a cancel button on them (normally it's icon is an 'X') if you press it should stop printing.

Though the other people have realistic ideas.. The sledgehammer technique does have the added benefit of stress relief and exercises. The only real downfall is the you do have to replace the dang thing..but you do feel better
