Under the Act of Settlement, Charles is the Heir Apparent to the throne, whether you happen to like the idea or not. That means that when the Queen dies, if he is still alive, he will ascend to the throne. After that, it will be the turn of Prince William and, if he fails to produce any heir, Prince Harry. I imagine that Prince Harry is quite relieved to think that all being well, he will probably be spared the burden of being King. He doesn't look as though he would enjoy spending part of each morning going through state papers and signing endless documents.

Prince Charles of Wales is next in line after Queen Elizabeth II. Prince William of Wales is second in line followed by his brother Prince Harry of Wales who is third in line.
The throne always goes to the monarch's first-born male heir as established by the line of succession act.
As for Prince William, I don't think he is ready to be King as he is still young and inexperienced (engaging in royal duties), unlike Prince Charles who had spent almost half of his life preparing for his role as King.
Someday in the future, Prince William will be King and he will definitely be a great monarch since he has two great inspirations in life, his grandmother the Queen and his mother, the late Princess Diana of Wales.

Normally the oldest one, but Charles is in the running for it. I don't really follow the royal line, so I'm not sure how that happened that William was after Charles, but I'm sure someone else will say something about that. And yes, they can pass on the crown pre-death, but the Queen is holding on pretty dang tight to that crown that she's taken a liking to.

Ummm Her oldest son, Charles. this is the way it somewhat works with royalty, you already know...The oldest son, in virtually any united states of america, or oldest new child in some....yet, in the known international, the youngster of the present monarch is it. And, if Charles could desire to predecease his mom, then William, HIS first new child, is next in line. this is all obtainable on Wikipedia.

Prince Charles.He is a direct descendant of the queen and mostly portrayed as the next in line.The british monarch is not run in a fashion where the queens sibling next to her is entroned.e.g the incumbent queen is a direct daughter of the late queen Elizabeth 1.

If Charles is alive I guess it'll have to be him but I reckon he should stand aside and let Wills be King, there is no way Camilla should be Queen, which she will be whatever they have said about it not being the case.
