Lots of good and some humorous answers...I can only counter with...whatever you do...DON'T try the old "Whooooops!" stunt. Not funny.
So I know this woman who had been married awhile and decided to finally try this.
She was a bit...uncomfortable...the following day...and finally at work, she had to tell her staff that she was not feeling well and had to go home.
She could barely keep a straight face when a male co-worker approached her the next day when she returned and said he had to go home because he must have what she had yesterday....

Although you should use your best judgment and wait for the exact moment, I think it is one of those things you just need to go for the conversation. Either wait until you've both had enough drinks and bring it up where you can always claim it was the alcohol or just bring it up out of the blue. She might find the conversation so amusing due to the randomness that she will actually hold that conversation with you.

If she isn't an open person sexually, you can pretty much forget it.
If she is open sexually, try watching a porn together and asking if she wants to try the same things the actors are doing. A bottle of wine won't hurt either.
But if you are going to try it, make sure you know what you are doing. Make sure you have lubrication and you choose the right position (not from behind) so it is as comfortable as possible for her. Believe it or not missionary position is the easiest position to try first. If it isn't comfortable for her, I can guarantee you won't get her to try it again!

A lot of women do not like that because it is painful and sounds pretty gross to them. I would just get in the throws of passion and start massaging her back door. If she is into it then continue, just lube up well. If she pushes you away...let it go. It is not worth pushing the issue.

Listen to John S. Don't ask her, it may gross her out or sound painful. Just try the tip of your finger while you are giving her oral and she is really wet and horny. I would bring it up slowly and see how she takes it. Also during sex I would try and play with it a little and if she tells you to stop then you won't ever get it.
