From NC to Kentucky:

Take I-40 west past Asheville to Knoxville

Once in Knoxville, take I-640 west for about five miles

Then, take I-75 north to Lexington. It will be about 170 miles from Knoxville to Lexington. (Of course, only you know where you're going in Kentucky, so I'll leave that up to you.)

Side notes:
-If you're going to Louisville, I-75 north will merge onto I-64 west in Lexington at exit 118; take I-64 west.
-If you're going to Mt. Sterling or Ashland, take I-64 east at exit 111 in Lexington.
-If you're going to Northern Kentucky, well, just keep truckin' on I-75.
-If you're going to Hazard or Pikeville, take the Hal Rogers Parkway east at exit 41 in London.
-If you're going to Somerset or Bowling Green, take KY 80 west at exit 41 in London. It's about 30 miles west from there to Somerset and, if you're going to BG, then KY 80 becomes the Louie Nunn Cumberland Parkway at Somerset.

75 to 81 to 40 appears to be the only way to avoid W. Virginia. But it might not be the best route to take. It depends upon your starting and ending points.

Not knowing where you are in KY or where you are heading in North Carolina, here are the main roads you would take without going through West Virginia:

I-75 South to around Knoxville
I-64 E towards Asheville
I-40 East

Just map quest it. I mapped it from my daughter's home in Newport, KY to my home in Claremont, NC.
