The movie I am pretty sure was called "El Dorado" with John Wayne saying this to Dean Martin. And James Caan was in this movie along with them. I think it was made in the mid 60's. This was a good question and yes it was the funniest saying in the movie considering John Wayne was usually not one to say something funny.

That was a good line. I had to get my hubby on this. He's got
about half of John Waynes' films. It's from El Dorado, with
Robt. Mitchum, and James Caan.
Seems it was a remake from Rio Bravo, which starred
Dean Martin, Ricky Nelson, Ward Bond, and Walter Brennan.
I think my favorite John Wayne film, was McClintock, with
Maureen Ohara. To me they were the perfect onscreen
match. I love the scene where they slide down in the mud
to the mud bath below, in that movie. They also were the
perfect match, in The Quiet Man, filmed in Ireland. Boy,
they sure don't make them like that anymore.
