It's a way of separating chemicals, compounds, molecules based usually on their size by causing them to migrate through a matrix.

The simplest is paper chromatography. You put a spot of your chemical mixture on a piece of absorbant paper, then stick the end of the paper in a liquid solution and allow it to wick up the paper. It will pick up the chemicals in the spot as it moves along and they will wind up being separated based on their size, since larger compounds can't get through the paper matrix as fast as smaller.

Electrophoresis, which is what's usually done to separate DNA molecules, is a similar idea, but the stuff migrates through a gelatin matrix on an electric current.
