What kind of sand? For pool filter sand (diatomaceous earth), about 60 pounds would be sufficient. Certain sands are more dense than others and will vary regarding how many pounds you need. I would suggest diatomaceous earth (it is GREAT for live plants and the fish love it too). To siphon it, use just the hose (not the "head" part that comes with it) and hover it just above the sand. This will give you about 1.5-2 inches of sand.
What do you need to glue? For any aquarium "gluing", use aquarium silicone. It is not toxic and will not harm your fish in any way. Email me if you have any questions.

What do you mean, glue? For what? If it's to seal the tank, I wouldn't bother because if there's a chance the tank has a leak you should be worried.
And about two to three inches of sand should suffice.

You do not use glue in a fish tank, the piosons will kill the fish.
I wouldn't recommend sand either, you will end up with a cloudy water when the fish swim about and churn it all up. You will never see your fish again.

Use as much sand as you want, unless you have an undergravel, then just enough to cover it. Glue? For what? If it's a crack then use a non toxic silicone.

Gravel should be about one inch deep and the only safe glue for aquariums is clear silicone.
