It sounds like you have a home page hijacker program installed. Download Adaware or another adware removal program and scan your computer.

It may have been set via policy. You can try deleting a registry key to re-enable it.
Start -> Run - Regedit
Browse through the tree view down to the Control Panel key:
HKEY_LOCAL_MACHINE
\Software
\Policies
\Microsoft
\Internet Explorer
\Control Panel
Ensure Control Panel is selected in the left pane. Look for a key in the right pane called "HomePage". If you find it, delete it.
Do the same for HKEY_CURRENT_USER
\Software
\Policies
\Microsoft
\Internet Explorer
\Control Panel
Log off or restart your PC and then try to change your home page.

I dont think it is possible to disable it. Even if your temporary files are enabled or history is enabled, it doesnt make any differents

is the grenral tab where you set the homepage still there ? if not go to this site download the reg fil and run it ... http://www.kellys-korner-xp.com/regs_edits/iegentabs.reg
this will put the genral tab back....
if the tab is there and the home page has been set to a specific site and locked then do this
click start run and type in regedit then press enter
go to this string HKEY_CURRENT_USER\Software\Policies\Microsoft\Inte rnet Explorer\Control Panel\Homepage once here rigth click on the word hompage and select modify...change the value data to 0 click ok...this will unlock the hompage..... remember 0 to open hompage and 1 to lock it...

Usually when certain functions are "disabled" it's because the administrator has placed password protection on changing any settings.

i didn't know it could be disabled. it sounds to me like a virus or some spyware called a brower hijacker. scan your computer for viruses and spyware
