You have a clog in the main line directly below the toilet in question. You should be able to run a 100 foot snake or rooter down the vent stack on the roof to the clog, Or pull the toilet on the 2nd floor and go through that drain pipe to the clog. If you work from below it, be prepared for a surprise visit of all that backed up water.

You have a plug in the first floor. Take a "plumbers helper" a toilet plunger and try to plung the toilet, if it goes down fine, flush it a few times, then try the second floor again, if it goes down, fine, then go to the third floor, if that one is plugged, then that's where the plug is. Plung it, if it won't clear you might have to call a plumber.

You do have a clog somewhere. Try holding a plunger down ( in the bowl it come out ) very hard in the hole. It should stop most of the water from coming in. I had to do that once when we had a clog.
Since the water is going up, the problem should be down. So, my suggestion would be to go to your local hardware and rent a "snake". It will get the clog loose for you. It's all mechanical too. Not to hard to do.

between the 1st and 2nd floor you have a blockage in the line,only cure pull toilet and snake out,wish i could be of more help.

the drain is plugged...u can use a snake to free it or the plunger or a chemical agent like liquid plumber...good luck

Is this an older home? You may need to have your drain rotted out by a professional. It is not expensive, should be $100.00 or less. I would suggest that 1st!
