Your machine is infected. Pop-ups are mostly caused by adware, spyware, and other malicious and unwanted software and using pop-up blocker won't help an infected PC. Since no single program can detect and eradicate all spyware, I recommend using multiple anti-spyware programs. Ad-aware and Spybot are among the best (they are free), but feel free to use additional programs if you have a nasty infection.
Running these programs in Safe Mode (tap F8 as the computer boots up and choose to boot to safe mode) can be more effective.
If you do not have an updated anti-virus program always running, you need to get one. Install only one, but you can get AVG, Avast! or AntiVir for free.
I also recommend using FireFox, it's so much better than Internet Explorer.
Since you stated you have run anti-spyware programs already, I must inform you to run Hijackthis. It's the most powerful one, but you must be careful not to remove anything without checking with the online community as it will it will find both legitimate and malicious software.

Difficult to say without knowing what you are you using for AV/Firewall and anti-spyware but
1 Get rid of all spyware that is aready on your computer. There are plenty of freebies out there. I find a combination of Webroot's Window Washer, RegCleaner and ScanSpyware does the job. You may need to run them all several times.
2. Get rid of Internet Explorer and Windows Media Player as far as you can, without destroying your operating system. Real Player is another one that dumps crap on you hard drive. Use non-Microshit aligned programmes such as Media Player Classic and Mozilla Firefox.
If all else fails then you need to back up the data n your hard drive and re-install. Best move I ever made on my computers was to get rid of (or at least suppress! Very difficult to get rid of it entirely without using another operating system) the abomination that is Internet Explorer. Or, as another poster suggests, you could use a different operating system based on Linux.
