The bacteria are dead in a stained preparation, and the bacteria are free to move around in the hanging drop. I'd prefer the stained prep for morphology and the hanging drop for motility.

In a stained prep, you usually heat fix the smear to the slide, so you kill the bacteria.

In a hanging drop (which by the way are difficult to do and view in my opinion) shows live baceria which are (or are not) motile.
