we had a reservation at the Comfort Suites and cancelled, however, we have lost the cancellation # and the hotel is charging us for the full cost of the room. do we have any recourse?

Nope. They gave you that cancellation number for a reason...it was your "receipt" that they accepted it and didn't charge you.

Without that number, you have to pay.

Probably not, if you lost the cancellation number - you can't prove that you cancelled, so are most likely stuck for the payment.

You can contact your credit card company and tell them about the dispute. They are usually good about helping and will hold off payment until it's resolved. Unfortunately it sounds like you are the one that needs to be able to prove you canceled.
