Sound card might make a difference.

The speakers will make a difference. But the real difference is in the sound card.

Laptops use sound cards built into the motherboard. Desktop can as well but they have the ability to have one installed and the one's that you have to install into your machine are much better than anything that is built in.

firstly you got to check the sound card, its diffrent in laptop and desktop,
second is laptop power is limited, so your speaker wont get much power to have a loud or good quality sound

Not unless the desktop is fitted with a decent soundcard like a SoundBlaster Live or similar.
Otherwise they all seem to use the same crappy Realtek or CMedia onboard sound.

well if its using the exact same thing laptop would be better since the speakers would be nearer to you rather than if its on your desktop and it is located on the floor away from you

depends.
if both are using crappy integrated audio, then they'll both sound bad.

the desktop at least will have the room for a dedicated soundcard, in which case, the desktop will provide better sound.

It is all down to purchasing a good sound card and of course setting it properly and finally getting a woofer, buffer, and roofer lol
