If it's across a huge resistance, then yes. The input impedance for most meters is in the megaohms. Thus if you put it across a megaohm R in a circuit, you've halved the resistance in that circuit.

Depends on the quality of the DMM you're using. Really good ones don't influence the circuit under test to any significant degree at all.
