If the position of an object is zero, does its speed have to be zero?

The object, if it is not moving, the speed is 0. It always has inertia though.

No. It could be moving with some non-zero velocity, and for an instant move over the 0 position. It doesnt necessarily have to be at rest when it is on this position.
